    Epidermis recreation in spongy-like hydrogels: New opportunities to explore epidermis-like analogues

    [Excerpt] On the road to successfully achieving skin regeneration, 3D matrices/scaffolds that provide the adequate physico-chemical and biological cues to recreate the ideal healing environment are believed to be a key element [1], [2] and [3]. Numerous polymeric matrices derived from both natural [4] and [5] and synthetic [6], [7] and [8] sources have been used as cellular supports; nowadays, fewer matrices are simple carriers, and more and more are ECM analogues that can actively participate in the healing process. Therefore, the attractive characteristics of hydrogels, such as high water content, tunable elasticity and facilitated mass transportation, have made them excellent materials to mimic cells’ native environment [9]. Moreover, their hygroscopic nature [10] and possibility of attaining soft tissues-like mechanical properties mean they have potential for exploitation as wound healing promoters [11], [12], [13] and [14]. Nonetheless, hydrogels lack natural cell adhesion sites [15], which limits the maximization of their potential in the recreation of the cell niche. This issue has been tackled through the use of a range of sophisticated approaches to decorate the hydrogels with adhesion sequences such as arginine-glycine-aspartic acid (RGD) derived from fibronectin [16], [17] and [18], and tyrosine-isoleucine-glycine-serine-arginine (YIGSR) derived from laminin [18] and [19], which not only aim to modulate cell adhesion, but also influencing cell fate and survival [18].     Stability of coordination polymers in water: state of the art and towards a methodology for nonporous materials

    A mini review on the study concerning water stability of coordination polymers (CPs) is presented. Next, following the procedure proposed recently by Gelfand and Shimizu (Dalton Trans 45:3668-3678, 2016) the stability of three cysteine (Cys)containing CPs is investigated. The stability of studied CPs decreases in the order: Zn(Cys)(2)>Mg(Cys)(2)>Ca(Cys)(2) H2O. For the latternever reported before, the structure is additionally determined and it is proved that water is located in the first coordination sphere. It is shown that for nonporous CPs, in contrast to the porous ones, the immersion in water at 20 degrees C is more drastic for studied solids than the harsh humid conditions (80 degrees C at 90% R.H.). Finally all materials are assigned to the hydrolytic stability groups and it is concluded that the stability of studied CPs correlates well with the standard reduction potentials. This leads to the conclusion that the application of more inert metal as a node causes larger stability of studied CPs

    Damage threshold of CuCrFeTiV high entropy alloys for nuclear fusion reactors

    A CuCrFeTiV high entropy alloy was prepared and irradiated with swift heavy ions in order to check its adequacy for use as a thermal barrier in future nuclear fusion reactors. The alloy was prepared from the elemental powders by ball milling, followed by consolidation by spark plasma sintering at 1178 K and 65 MPa. The samples were then irradiated at room temperature with 300 keV Ar+ ions with fluences in the 3 × 1015 to 3 × 1018 Ar+/cm2 range to mimic neutron-induced damage accumulation during a duty cycle of a fusion reactor. Structural changes were investigated by X-ray diffraction, and scanning electron microscopy and scanning transmission electron microscopy, both coupled with X-ray energy dispersive spectroscopy. Surface irradiation damage was detected for high fluences (3 × 1018 Ar+/cm2) with formation of blisters of up to 1 μm in diameter. Cross-sectional scanning transmission electron microscopy showed the presence of intergranular cavities only in the sample irradiated with 3 × 1018 Ar+/cm2, while all irradiation experiments produced intragranular nanometric-sized bubbles with increased density for higher Ar+ fluence. The Williamson-Hall method revealed a decrease in the average crystallite size and an increase in residual strain with increasing fluence, consistent with the formation of Ar+ bubbles at the irradiated surface.publishedVersio

    Ruminal Degradability Of Agro-industrial Fruit Residues

    The aim of this study was to evaluate the chemical composition and ruminal degradability of the dry matter (DM), crude protein (CP) and neutral detergent fiber (NDF) of fruit residues. Three fistulated sheep were held collectively in a pen, and fed daily with the studied residues in a diet consisting of canarana grass (Echinochloa pyramidalis) and a concentrate of corn and soybeans. The animals were allowed an adjustment period of seven days. The residues were dried in the sun, crushed in a forage machine, sorted using a 4.0-mm sieve, and incubated for 3, 6, 12, 24, 48, 72, and 96 h using nonwoven bags (weight 60g/m2, 14 ×12 cm2). Chemical analyses of the residues were performed using a randomized block experimental design with split plots. The cherimoya and tamarind residues showed the highest concentrations of CP (12.66% and 11.79%) the ether extract of cherimoya residue was the highest at 22.30%stands out the sour soup residue. The cashew and guava residues showed the highest levels of lignin (22.13 and 18.34%). The effective degradability of DM for the pineapple and tamarind residues to a passage rate of 5%/h were 53.04% and 42.61%, respectively. The guava, cherimoya, and cashew residues showed lower values at 19.16%, 26.86%, and 29.21%, respectively. The cherimoya, guava and pineapple residues showed the highest values of potential degradability for CP at 87%, 81%, 86.02% and 90.94%, respectively, with an average effective degradability of 50.0% at the rate of 5%/h. The pineapple (35.38%) and tamarind residues (34.49%) showed higher values of the effective degradability of NDF at a passage rate of 5%/h. Among the studied residues, the pineapple residue showed the greatest potential for use in animal feed based on chemical composition and rates of degradability.37127929

    Hypothalamic Endoplasmic Reticulum Stress Of Overtrained Mice After Recovery

    knowing the relationship between endoplasmic reticulum (ER) stress and inflammation and based on the fact that downhill running-based overtraining (OT) model increases hypothalamus levels of some pro-inflammatory cytokines, we verified the effects of three OT protocols on the levels of BiP, pIRE-1 (Ser734), pPERK (Thr981), pelF2alpha (Ser52), ATF-6 and GRP-94 proteins in the mouse hypothalamus after two weeks of recovery. Methods: the mice were randomized into control (CT), overtrained by downhill running (OTR/down), overtrained by uphill running (OTR/up) and overtrained by running without inclination (OTR) groups. After 2-week total recovery period (i.e., week 10), hypothalamus was removed and used for immunoblotting. Results: The OTR/down group exhibited high levels of BiP and ATF6. The other OT protocols showed higher levels of pPERK (Th981) and pelf-2alpha (Ser52) when compared with the CT group. Conclusion: The current results suggest that after a 2-week total recovery period, the overtrained groups increased partially their ER stress protein levels, but without hypothalamic inflammation, which characterizes a physiological condition related to an adaptation mechanism.2

    This research was conducted at the Experimental Station of Piracicaba, São Paulo, Brazil. Six sugarcane cultivars were studied with the main purpose of comparing the percentage of infested stalks and the intensity of infestation by the borer - rot complex. The cultivars were planted in April of 1985 and the evaluations were made in May, July and September of 1986. The results indicated that there were no differences between the internal and external percentages of infested stalks. The internal and external intensities of infestation presented a positive and highly significant linear correlation. On the other hand, a significant correlation between the percentage of infested stalks and the intensity of infestation was not always found. The highest percentage of infested internodes was observed at the basal region of the stalk and the lowest at the apical one.O presente trabalho foi desenvolvido na Estação Experimental de Piracicaba, SP, utilizando-se seis cultivares de cana-de-açúcar, com o objetivo de comparar a porcentagem de colmos infestados e a intensidade de infestação como técnicas para avaliar o ataque pelo complexo broca-podridões. As cultivares foram plantadas em abril de 1985, sendo as avaliações realizadas nos meses de maio, julho e setembro do ano seguinte. Com base nos resultados obtidos, constatou-se que não há diferenças entre as porcentagens de colmos infestados externa e internamente pelo complexo broca-podridões. A intensidade de infestação interna foi maior que a externa, verificando-se entre estes parâmetros uma correlação linear positiva e altamente significativa. Nem sempre se constatou correlação significativa entre porcentagem de colmos infestados e intensidade de infestação. In order to study the effects of water stress on the sweet pepper crop, an experiment was conducted in greenhouse. The treatments consisted of soil water content (100%, 85%, 70% and 55% of moisture in the soil field capacity) in a randomized block design with 6 replicates where the experimental units consisted of 16 plants. During the experiment (47, 62, 77 and 92 days after planting) levels of macronutrients (N, P, K, Ca, Mg and S) and micronutrients (B, Cu, Fe, Mn and Zn) present in each plant fraction and the dry mass of plants for the accumulation of these determination were determined. At harvest the number, width and length of fruits from each plant were obtained. Variables were subjected to Tukey test at 5% probability. The water content in the soil interfered significantly with productive variables. Subjects and methods: Samples of nineteen 46,XY healthy young men and five patients with disorders of sex development (DSD), four 45,X/46,XY and one 46,XY were used. FISH analysis with X and Y specific probes on interphase nuclei from blood lymphocytes and buccal epithelium were analyzed to investigate the proportion of nuclei containing only the signal of the X chromosome. Results: The frequency of nuclei containing only the X signal in the two tissues of healthy men did not differ (p = 0.69). In all patients with DSD this frequency was significantly higher, and there was no difference between the two tissues (p = 0.38), either.
